German Gisdol has trained Hoffenheim, SV Hamburg and Cologne in the past. The 52-year-old succeeds Serbian Marko Nikolic whose contract was terminated earlier in the week.
Lokomotiv is fifth in the Russian Premier League, six points behind leaders and defending champions Zenit St Petersburg.
Gisdol is the second German coach of a Moscow club, with Sandro Schwarz in charge of Spartak Moscow since 2020.
